Development of multiplex PCR for Lr21 and Pm4b resistance genes detection in common wheat (Triticum aestivum L.)

Abstract

Leaf rust caused by Puccinia triticina and powdery mildew Blumeria graminis are considered to be one of the most significant diseases of common wheat (Triticum aestivum L.). The most efficient method of these diseases control and their effects limitation is breeding and cultivation of cultivars characterized by resistance to these pathogens. The aims of presented study were identification of leaf rust – Lr21 and powdery mildew – Pm4b resistance genes in Polish wheat cultivars, as well as development of multiplex PCR useful for simultaneous identification of these genes. The plant material of present studies were 30 Polish common wheat cultivars. Extracted DNA was amplified using in single reaction primers specific for both analysed genes. On the basis of the conducted research it was shown that the developed multiplex PCR is a technique useful for simultaneous identification of Lr21 and Pm4b genes. Multiplex PCR could be used in resistance breeding of common wheat supported by molecular markers for identification of these genes.
