Application of RAPD linkage map for identification of sprouting resistance genes in rye (Secale cereale L.)

Abstract

The aim of this paper was the construction of RAPD linkage map based on the intercross S120 × S76 and localization of QTLs underlying sprouting resistance. 876 10-mer primers were used for detection of inter-line polymorphism. 48 markers were found which segregated in F2 progeny according to a single gene model. The map constructed using these markers consists of ten linkage groups spanning the distance of 248.8 cM and covering about 20% of rye genome. Among the mapped markers three were common with those found earlier on the map constructed using DS2 × RXL10 intercross. A low number of common markers for the two maps precluded chromosomal identification of the newly obtained linkage groups. Two QTLs controlling sprouting resistance were identified within two different linkage groups on the S120 × S76 map.
