Analysis of growth and development of blue honeysuckle (Lonicera caerulea L. var. kamtschatica Sevast.) seedlings in in vitro culture

Abstract

The aim of the study was to determine the suitability of in vitro culture for the selection of seedlings derived from two blue honeysuckle cultivars ‘Atut’ and ‘Czelabinka’. The usefulness of in vitro culture for genetic and breeding work within this species was assessed on the basis of an analysis of growth and development of seedlings grown on medium Murashige and Skoog (MS) supplemented with cytokinins. The addition of this group of growth regulators was to assist in removing endogenous cytokinin apical bud dominance and thereby stimulate the shoots of seedlings for propagation. Two types of cytokines – 6-benzyladenine (BA) and kinetin (KIN) – at the concentrations of 4 and 2 mg · dm-3, respectively, were applied. During the culture, differentiation was observed in the growth and development of seedlings in the two populations of progeny. Shoots longer than 15 mm accounted for more than 60% in both populations. Among seedlings of cultivar ‘Atut’, the highest contribution of shoots with 5–10 leaves was observed on the MS medium supplemented with kinetin, while in the seedling population of cultivar ‘Czelabinka’ – on the medium supplemented with BA. The roots produced during the culture by both seedling populations showed various degrees of development. The seedlings of cultivar ‘Atut’ produced a greater number of roots, but the roots were short, while the seedlings of cultivar ‘Czelabinka’ produced fewer roots reaching the length of more than 5.0 mm. The obtained results may arise from genetic variation of the segregating progeny populations of seedlings, which would be due to the allogamy of the species. The studies are contributory and only indicate the potential use of in vitro selection within the species. Therefore, further experimental work should be carried out to determine the optimal conditions for efficient selection and breeding of blue honeysuckle in in vitro culture.
